"Dopo la partenza" means "after the departure". Probably means that the postage was paid after the departure (of the stagecoach ?).
"Il sindaco del municipio di Osimo" means "The mayor of the municipality of Osimo". The handwritten signature (of the mayor or his secretary ?) is illegible. The letter is addressed to the mayor of Macerata, in the same Province.
"Vetus Auximon" (Old Osimo) is the Latin name of Osimo. The round cancellation represents the city seal.
Osimo is a small city on the Adriatic coast of Italy, near Ancona in the Marche Province.

The words "Dopo la partenza" mean "after the departure" and nothing else. They do not say anything about postage. If you are on an Italian plane, you may be told that a meal will be served "dopo la partenza", but I am going to search the internet to try finding out why this cancellation was used.

This Italian link explains that the stamp "dopo la partenza" was put on letters which arrived at the post office after the post stagecoach had left, in order to explain why the letter then took longer to arrive.
Thus, it does not have any significance regarding postage.

"per il timbro DOPO LA PARTENZA bisogna spezzare una lancia a favore dell'Amministrazione Postale Granducale poi Sabauda, che pare sia stata una delle migliori per l'epoca, per il ramificato servizio e la funzionalità nel trattare le corrispondenze, ebbene questo timbro lineare stampatello DOPO LA PARTENZA veniva apposto sulle lettere consegnate all'ufficio postale dopo che il corriere o diligenza che si voglia era già partita ovvero al di fuori dell'orario stabilito, allora cosa succedeva siccome la lettera era stata consegnata per quella data, ma fuori tempo, gli veniva applicato quella dicitura per giustificare l'inoltro per la spedizione con la corriera successiva e quindi la consegna al destinatario ritardata rispetto alla data prevista nel caso contrario,............................. mi è capitato spesso di leggere all'interno delle lettere frasi del tipo.......Devo frettolosamente completare la lettera altrimenti non faccio in tempo con la consegna giornaliera perchè è prossima a partire la diligenza..... ecco quindi nei casi dove questo purtroppo a causa del ritardo di consegna del mittente succedeva, alla missiva gli veniva apposto questo timbro di giustificazione.........................anche perchè a quel tempo in base a cosa avevo scritto, aspettavano a tempo debito la risposta e in quel caso non l'avevano per il tempo stabilito a seconda del percorso ma in ritardo e quando la ricevevano capivano il perchè del ritardo da quel timbro.........................scherzavano poco all'epoca, non si sgarrava, spedire costava a quel tempo, ma le cose erano ben fatte, le lettere all'interno della toscana se consegnate al tempo debito erano consegnate nello stesso giorno"
It is a bit lengthy and repetitive, but it's funny to read that the letter writer was worried to miss the departure of the stagecoach (diligenza) ... and then missed it !
